Samples are prepared by mixing with an extraction solution(or just centrifuging), blending and filtering. The extract is then applied to the Lactoferrin immunoaffinity column bound with Heparin.At this stage, the Lactoferrin bind to the Heparin on the column. The column is then washed with water to remove the impurities. By passing Elution Solution through the column, the Lactoferrin is removed from the IAC. This Elution Solution can then be injected into HPLC system.
A simple and efficient extraction and purification procedure for Lactoferrin was developed by means of the immunoaffinity column (IAC - SEP ® Heparin) as a cleanup tool. This method do not use toxic solvents such as chloroform and methylene chloride. Lactoferrin content in milk, milk powder are cleaned up by IAC and determined by HPLC. It is a fast, simple, safe and highly accurate method for quantitatively measuring Lactoferrin.
3.0 Storage Conditions
The affinity column should be stored at 2-8°C and must not be frozen. The shelf life is 18 months. It is recommended for use at room temperature (18-30°C).
