1. BACKGROUND
Aflatoxin is a naturally occurring mycotoxin produced by two types of mold: Aspergillus flavus and Aspergillus parasiticus. AflatoxinB1, B2, G1 and G2 can be found mainly in cereals, corn, cotton seed, some nuts and in spices. Aflatoxins can cause liver disease in animals and may cause decreased production (milk, eggs, animal weight, etc). Aflatoxin B1 is a potent human carcinogen, and may contribute to human liver cancer. Most controlling government agencies worldwide have regulations regarding the amount of aflatoxins allowable in human and animal foodstuffs. Accurate and rapid determination of the presence of aflatoxin in commodities is of very importance.
2. PURPOSE
This handbook establishes procedures for determining total aflatoxin in grain, processed grain products and feed. The test is a direct competitive ELISA that provides exact concentrations in parts per billion (ppb). Free toxin in the sample and controls competes with enzyme-labeled toxin (conjugate) for the antibody binding sites. After a wash step, substrate reacts with the bound enzyme conjugate to produce blue color. A stop solution is then add which changes the color from blue to yellow. The intensity of the color is inversely proportional to the concentration of aflatoxins in the samples and standards.
